Abstract
Single cell analysis has recently emerged as an important field of biomedical research. It is now clear that heterogeneity in complex biological systems is correlated to changes in biological function and disease processes. A variety of micro/nano probes were developed to enable investigation of cells properties such as membrane stiffness, pH value. Nevertheless, very few designs were focused on single cell metabolism studies, and the existing approaches suffer from fundamental defects and need to be improved.
In the present research, we propose a single cell respirometer that can be used to investigate cell metabolism for a long period of time at single cell level. Specifically, this tool is designed to sense cell oxygen consumption, ion exchanges and metabolites flux. Data acquired with this proposed device will provide valuable assistance on applications like cell metabolism study, cancer diagnose, or therapy evaluations.
One critical design of the proposed device is a micro perfusion platform, which allowed us to isolate one single cell in a small volume of a constant environment. We will evaluate this perfusion system by both simulation and experimental method. With this micro-perfusion system as a platform, we will design probes for cell metabolic function sensing. We will experimentally evaluate these sensors sensitivity, response time and their accuracy. Finally, this device will be used to investigate the metabolism of metastatic and normal prostate cells. |
