Abstract:
Osteoarthritis (OA) is characterized by the irreversible breakdown of articular cartilage and the loss of joint load-bearing function. It affects nearly 27 million people in the United States. The effective early diagnosis and functional cure for OA is currently hindered by the lack of understanding of how molecular activities in cartilage are linked to its macroscopic function. This renders it important to study the nanoscale building blocks of cartilage extracellular matrix (ECM). In this study, we will reveal the nanoscale structure-mechanics principles of the ECM in the function and disease progression of cartilaginous tissue. Specifically, roles of decorin and biglycan in cartilage nanostructure and nanomechanics during skeletal maturation and nanomechanical phenotype of post-traumatic osteoarthritis (PTOA) after destabilization of medial meniscus (DMM) will be studied.
