The Drexel Advanced SPR Biosensor Resource is housed in the Drexel College of Medicine (NCB11302, Center City Philadelphia). SPR affords:

SPR biosensors measure molecular interactions in real time. Several other methodologies determine affinity of complex formation, such as ELISA, analytical ultracentrifugation, equilibrium chromatography, and isothermal titration calorimetry, using absorbance, fluorescence, and temperature change as reporters of binding. Based on the integration of a controlled, fluidic system and the detection of refractive index changes at a surface, SPR reveals the dynamics of complex formation and dissociation, which is critical in understanding the function of biomolecules at high resolution.
Biosketch:
Gabriela Canziani, PhD, is the Drexel-SKCC/TJU SPR Biosensor Facility Manager, under the direction of Dr. Irwin Chaiken. The facility was established in 2020 following the NIH funded purchase of an S200 high sensitivity Biacore biosensor. Dr. Canziani has been an active participant in the evolution of SPR systems since their introduction as a biophysical tool and has become expert in the study of molecular recognition, a field of biomolecular research that explores the mechanisms of non-covalent binding between two or more molecules, and their correlation with complementary biophysical technologies to identify and explore the mechanism of specific binding interactions.
With more than 25 years of experience in delivering molecular interaction analysis core services to investigators in academia, government institutions, and as lead scientist in a protein biophysical characterization resource in the Pharma/Biotech industry, Dr. Canziani has been part of basic research and R&D for more than 20 extracellular and intracellular therapeutic targets, and in the definition of four lead therapeutics and two new molecular entities in the pharma pipeline. In addition, she has supported discovery by medium throughput compound screening followed by analysis of selected hits using other biophysical methods, such as ITC, Thermal Shift, Static and Dynamic Light Scattering (SLS and DLS), SEC, and Differential Scanning Calorimetry (DSC). She has co-authored 40 papers, a patent, has earned research funding, and has helped instrument users to obtain funding. |
