Contemporary culture most often places value on authoritative individual (master) voices—when often those voices rely upon the work of others. In an attempt to de-center the focus on solo auteurs as preferred makers, Assemblage looks to highlight works by collectives or collaborative projects made by several artists. All of the artists featured in the show move in and out of disciplines, cities, and venues—defying categorization or genre, much like the show’s curator. Curated by Maori Karmael Holmes, Founder and Artistic Director of the Blackstar Film Festival.

Exhibition from March 29, 2019 through July 26, 2019. The late Matt Phillips (1927–2017) was a painter, printmaker and art educator. He was born in New York and studied literature at the University of Chicago where he earned his master’s degree. He first studied art at the Barnes Foundation in Merion, Pennsylvania, and traveled throughout Europe studying painting. He taught art in Paris from 1962 to 1964. When he returned to the United States, he taught at Bard College in upstate New York for 27 years.

This exhibition focuses on how Phillips, who is best known for his monotypes, and who often referred to himself as a “painter-poet,” found inspiration in several areas, notably poetry, nature, the female form and his world travels. His concentration on the medium of monotyping, helped to reintroduce contemporary artists to a technique that had been favored by nineteenth-century artists.

This exhibition was curated by Joseph O’Kane ’18 as a museum leadership graduate practicum project. Special thanks are owed to Arthur L. Frank, MD, PhD, professor and chair emeritus of the Department of Environmental and Occupational Health at the Dornsife School of Public Health, and his wife Joanne B. Frank, for their sponsorship and the use of their collection. Without their generosity, this exhibit would not have been possible.

John Chapel, Jr. ’67, taken at his home in Kiawah Island, South Carolina, throughout the region and across the world, over twenty-nine countries, from trips with his wife, Jinnie. Chapel received his first camera in the sixth or seventh grade, but it wasn’t until he and his wife began traveling that Chapel became a more avid photographer. Now he is part of the Kiawah Camera Club and works alongside and learns from other photographers to hone his skills. The subject matter of the photographs ranges from flowers to animals to architecture, something Chapel is particularly drawn to due in part from his engineering courses at Drexel University.
Between 2008 and 2014, John and Jinnie Chapel have donated over $3.5 million to the Drexel LeBow College of Business, including a $2.5 million gift endowing the dean’s chair. The Chapels also support Drexel’s Department of Athletics by establishing the John and Jinnie Chapel ACHIEVE Center and Dragon Lounge, where all of the University’s current Division I varsity student-athletes can receive support in academic, leadership, career and personal development, as well as engage in civic engagement opportunities.

Chapel is continuing his generosity to Drexel by making the photographs in this exhibition available for purchase, with all proceeds going to the University.

School of Biomedical Engineering, Science and Health Systems
Drexel University

Details:
Congestive heart failure (CHF) is a progressive and debilitating disease that affects millions of adults and children worldwide. Hundreds of thousands of new cases of CHF are diagnosed each year in the U.S., and this high volume of patients costs the healthcare industry tens of billions annually. The inadequate number of donor hearts and limited long-term effectiveness of pharmacologic treatment necessitate the development and use of mechanical circulatory support (MCS) therapies. Clinical utilization of MCS devices has demonstrated that patients derive substantial survival and quality of life benefits from short and long-term MCS. Only two total artificial hearts (TAHs) are approved for implantation in the U.S., and the implementation of TAHs in the treatment of patients with CHF has increased more than 3 fold in the last 6 years. To address the limitations of current technology and to provide a new and novel therapeutic alternative, we are developing an innovative, hybrid-design, continuous flow, implantable, magnetically levitated TAH (Dragon Heart).

The Dragon Heart is designed to provide cardiovascular support to pediatric and adult patients. This compact medical device has a target diameter and height of 70 mm by 50 mm, respectively; it is designed to generate the physiologic pressures and flows necessary to support CHF patients. This TAH has only 2 moving parts: an axial flow impeller to drive blood flow through the pulmonary circulation and a centrifugal flow impeller to drive blood flow through the systemic circulation. This device utilizes the latest generation of magnetic bearing technology to levitate the impellers, thus enabling an operational lifespan of 15 years. The design incorporates wider clearances between the rotating impellers and pump housing. Larger clearances lead to lower fluid shear stresses, hence mitigating the risk of thrombosis and hemolysis. This design avoids the use of mechanical or biologic valves and has an antithrombogenic coating applied to blood-contacting surfaces, thus further reducing the risk of thrombosis. It integrates state-of-the-art monitoring with Wifi-based sensors to report operational status and produces both continuous and pulsatile blood flow. A transcutaneous energy transfer system using wireless technology will be incorporated to supply power to the device and to eliminate the percutaneous driveline that protrudes from the abdomen, which is a major source of infection for patients who are supported by current MCS technology. This dissertation research concentrated on advancing the development of the axial and centrifugal blood pumps that constitute the core components of the Dragon Heart.

Three substantial design phases were completed. The pump geometries (axial and centrifugal) were established using advanced pump design equations, 3D modeling and pump design software, and available literature on blood pumps. ANSYS computational fluid dynamic software was used to evaluate the pressure flow generation, irregular blood flow velocities and patterns, estimated power consumption, radial and axial fluid forces on the rotor, fluid stress levels, blood cell residence time in the device, and blood damage predictions. The computational models served as the foundation by which prototype manufacturing was completed for hydraulic testing. Three hydraulic test rigs were constructed to test the pressure generation and levels of hemolysis in the prototypes. A blood analog solution of water and glycerin was utilized for the hydraulic experiments, and bovine blood was used for the repeated hemolytic studies.

During the design process, the axial and centrifugal pump designs were improved iteratively to reduce the size of the pumps without compromising pump performance. This process resulted in a small device size of 67 mm by 50 mm, surpassing the target goal. The simulations revealed that both pumps were able to generate adequate pressure and blood flow to support adult and pediatric patients. Computational results followed expected theoretically trends for pumps in general. Radial and axial fluid forces were estimated to be less than 0.4 N and 3 N, respectively. Maximum fluid stress levels remained less than 320 Pascal with maximum exposure times of less than .54 seconds. Blood damage models for several operating conditions indicated low damage levels of less than 2%. Estimated power consumption was determined to be less than 6 Watts.

Physical prototypes of the axial and centrifugal pump designs were manufactured for hydraulic and hemolytic testing. The experiments demonstrated that the Dragon Heart is capable of delivering the target blood flows of 1-5 LPM and pressure rises of greater than 15 mmHg for the pulmonary circulation and greater than 95 mmHg for the systemic circulation for acceptable rotational speeds. The centrifugal prototypes had a general trend of outperforming the computational predictions by 37.5%; while the axial flow prototypes showed a decreased pressure-flow range during testing as compared to the computational studies. The data trends of the hemolytic studies followed expectations with a linear increase in plasma-free hemoglobin levels, slow decrease in hematocrit, and a consistent maintenance of operational conditions of the pump during the experiments. The Normalized Index of Hemolysis levels, however, were higher than anticipated in comparison to clinically used MCS devices. This is likely due to the materials of construction of the prototypes and surface roughness factors, which will be addressed in future work.

This research and development effort is significant progress in the design of the Dragon Heart and represents a strong foundation by which to begin further optimization and development. The long-term goal of this research is to commercialize a novel, less expensive, more compact, low thrombus, and effective therapeutic alternative for adult and pediatric patients with CHF, thereby saving thousands of lives annually.

  Title:
Designing a Novel Immunomodulatory Surface Modification To Promote
Biomaterial-tissue Integration

Speaker:
Emily Beth Lurier, PhD Candidate
School of Biomedical Engineering, Science and Health Systems
Drexel University

Advisor:
Kara L. Spiller, PhD
Associate Professor
School of Biomedical Engineering, Science and Health Systems
Drexel University

Abstract:
The foreign body response (FBR) to implanted biomaterials provides a major challenge for successful biomaterial-tissue integration in the body. During the FBR, biomaterials are encapsulated in a collagenous, scar-like capsule that prevents the material from interacting with the surrounding tissue. Ultimately, the biomaterial will fail due to degradation and encapsulation, resulting in the removal and replacement of the implant. The primary culprit of the FBR is macrophages, the primary cell type of the inflammatory response. Macrophages exist on a spectrum of behaviors, typically transitioning from an “M1” pro-inflammatory to “M2” pro-healing phenotype in healthy wound healing. The M2 phenotype can be further broken down into interleukin-4 (IL4) stimulated “M2a” and IL10 stimulated “M2c” macrophages, although the differences between these phenotypes in biomaterial-tissue integration have not been fully elucidated. During the FBR, this phenotypic transition is halted, and macrophages exhibit chronic M1 behavior leading to fibrous capsule formation. While many biomaterial strategies to inhibit chronic M1 behavior and promote the natural M1-to-M2 phenotype switch have been investigated, there is currently no method to inhibit the FBR. Furthermore, methods to temporally control macrophage behavior to prevent chronic responses in either direction have not been successfully implemented. Therefore, the overarching goals of this work were to (1) design an affinity-based cytokine release system to modulate macrophage phenotype over time and (2) test the ability of the drug delivery system using biotin-avidin to temporally modulate macrophage phenotype both in vitro and in vivo.

First, an investigation into the M2c phenotype revealed that this phenotype may act at early stages of wound healing, primarily contributing to matrix remodeling. M2c macrophages were found to secrete high quantities of matrix degrading enzymes suggesting that this phenotype may not be beneficial in biomaterial-tissue integration. Comparatively, M2a macrophages have been previously shown to inhibit fibrous capsule formation and promote angiogenesis in the context of biomaterial-tissue integration. Therefore, the M2a polarizing cytokine IL4 was bound to Gelfoam scaffolds, used as a model biomaterial throughout, via the biotin-avidin affinity conjugation system.

The biotin avidin system has been previously shown to release proteins over time and has the potential for creating a loadable protein drug delivery system in vitro. However, the ability to modulate macrophage activation by controlling the parameters of the system over time have not been thoroughly investigated. The effects of modulating both the fold molar excess of biotin on the scaffold surface and altering the avidin variants were investigated. Increasing the fold molar excess (FME) of biotin on the scaffold conjugated with CaptAvidin, and not Streptavidin, was shown to decrease IL4 release over time. Additionally, in a similar manner, increasing the FME of biotin on the scaffold with CaptAvidin and IL4 decreased M2a activation in vitro. Scaffolds with low levels of biotinylation in vivo were shown to have the highest reduction in fibrous capsule formation compared to groups with no IL4, adsorbed IL4 or a high degree of biotinylation. However, differences in gene expression signatures of explants were minimal. A more thorough investigation is required to determine the in vivo effects of modified scaffolds on fibrosis and angiogenesis.

Overall, modifying scaffolds with biotin-avidin shows great potential in designing immunomodulatory drug delivery systems to control macrophage activation over time. Future studies should focus on utilizing the biotin-avidin system as a loadable cytokine release system to control the M1-to-M2 phenotypic transition which would have significant impact on treating pathologies where this transition is halted, like fibrosis and chronic wounds.

O’Donnell will recount highlights and (low points) of his recent trip to work in Amazonian flooded forest with a BCC natural history film crew. Through images and video he will reflect on the fascinating but poorly-studied ecology of a region of extensive flooded forest during the high-water season. Paleontological Expedition"

Logistical challenges are a big part of any fieldwork in remote parts of the earth. During December and January Dr. Daeschler and colleagues deployed into the interior of Antarctica to collect geological and paleontological data from the Middle Devonian-age Aztec Siltstone. High winds and drifting snow slowed down their team but ultimately they were successful in sampling a series of outcrops in the area around Deception Glacier. During the final week at Deception Glacier they were joined by two film makers as part of their effort to develop broader impacts.

With the battle over fact and fiction taking place between the American government, journalists, and general public, science writing is in a fragile state. How does a writer advocate for science during a period where science and its methods are routinely dismissed, derided, and discounted? What adjustments need to be made to demonstrate the veracity of findings, the interpretation of those findings, without further isolating those who need these findings most? This panel will address the events that have ushered us into this scientific quagmire, suggest ways science writing can adjust to address new needs, and contemplate science journalism's future.
