The show will connect the works of the collections and the work being created at Drexel to the history of photography and its uses. From tourist photography, to photojournalism, to museum documentation, photography’s importance to society and to Drexel will be explored, explained, and celebrated. Endothelial dysfunction, characterized by impaired nitric oxide (NO) bioavailability, arises in response to a variety of cardiovascular risk factors and precedes atherosclerosis. NO is produced by tight regulation of endothelial nitric oxide synthase (eNOS) activity in response to vasodilatory stimuli. This regulation of eNOS is mediated in part by store-operated calcium entry (SOCE) and Protein Kinase C (PKC), a promiscuous serine/threonine kinase. Calcium-dependent PKCbeta (PKCβ) and calcium-independent PKCeta (PKCη) have both been implicated in the regulation and dysfunction of endothelial responses to shear stress and agonists. This signaling pathway is thought to be compartmentalized in caveolae.

This study seeks to quantify the contribution of PKCη, PKCβ, Orai1 channels and transient receptor potential canonical (TRPC) channels in regulating calcium signaling and endothelial nitric oxide synthase (eNOS) activation after exposure of endothelial cells to ATP or shear stress. The role of Cav1 as a scaffold for the proteins that regulate this pathway was also investigated. Bovine and human aortic endothelial cells were stimulated in vitro under pharmacological inhibition of target PKCs, and calcium channels as well as following knockdown of Cav1. The participation of these proteins in calcium flux, eNOS phosphorylation and NO synthesis was assessed following stimulation with agonist or shear stress.

PKCη proved to be a robust regulator of agonist- and shear stress-induced eNOS activation, modulating calcium fluxes and tuning eNOS activity by multi-site phosphorylation. PKCβ showed modest influence in this pathway, promoting eNOS activation basally and in response to shear stress. Both PKC isozymes contributed to the constitutive and induced phosphorylation of eNOS. The observed PKC signaling architecture is intricate, recruiting Src to mediate a portion of PKCη’s control on calcium entry and eNOS phosphorylation. Elucidation of the importance of PKCη in this pathway was tempered by evidence of a single stimulus producing concurrent phosphorylation at ser1179 and thr497 which are antagonistic to eNOS activity. The peak and sustained ATP-induced calcium signal and the resulting eNOS activation were attenuated by inhibition of TRPC3, which was store operated. TRPC4 blockade reduced the transient peak in calcium concentration after ATP stimulation but did not significantly reduce eNOS activity. Simultaneous TRPC3 & 4 inhibition reduced flow-induced NO production via alterations in phosphorylation-mediated eNOS activity. Inhibition of TRPC1/6 or Orai1 failed to lower ATP-induced calcium entry or eNOS activation. Cav1 silencing reduced ATP-initiated eNOS activation.

For the first time, in a single species in vitro, shear stress- and ATP-stimulated NO production are shown to be differentially regulated by classical and novel PKCs. TRPC3 is shown to be store-operated in BAECs and HAECs and is the key regulator of ATP-induced eNOS activation, whereas flow stimulation also recruits TRPC4 into the pathway for the synthesis of NO in BAECs. This study furthers our understanding of the PKC and TRPC isozyme interplay that optimizes NO production and the importance of Cav1 localization of this pathway. These considerations will inform the ongoing design of drugs for the treatment of PKC-sensitive cardiovascular pathologies.

Frank Furness’ 1876 Centennial National Bank, located at the crossroads of Market and 32nd Street, marks the gateway to the Drexel University campus. Now the Paul Peck Alumni Center, it is home to a newly renovated gallery space designed to showcase Drexel’s rich and diverse collections. These collections, which embody the interdisciplinary collaborative spirit of Drexel academics mark Drexel as one of the great collecting universities in the United States.

Though widely known for innovation, entrepreneurship and civic engagement, Drexel has been a serious collecting institution from its very inception in 1891. In fact, it was the University’s forward thinking founder AJ Drexel, who provided the inspiration and resources to establish The Drexel Collection. The original collection grew and evolved over the years and now the University holds a series of impressive collections including the internationally renowned Academy of Natural Sciences of Drexel University, The Robert and Penny Fox Historic Costume Collection as well as The Legacy Center, which documents the role of women in American medicine, and the Archives at Hagerty Library.

The inaugural exhibition in the new gallery, curated by Cara Fry and Derek Gillman, is designed to provide a window into Drexel’s extraordinary collections which range from rare books to couture; from natural science to fine and applied arts. Every piece in the show is a treasure – including a Rittenhouse Orrery clock from 1873, a 1991 James Galanos evening gown, a 450 million year old slab of fossilized trilobites and a 16th century Paracelsus incunabulum. Also highlighted is an 1876 William Morris textile which serves as a fascinating case study for the way in which the objects in the Drexel collections invite interdisciplinary teaching and research and thereby provide historical context and contemporary relevance.

Exhibition from March 11, 2022 through March 2023. On March 11, the Anthony J. Drexel Picture Gallery in the Main Building will open A New Kind of House, a year-long exhibition celebrating eight years of Writers Room—a university-community literary arts program engaged in creative placemaking and art for social justice. The photography and writing of students and neighbors, displayed alongside and in conversation with pieces from The Drexel Collection, will engage in a rich dialogue about the changing face of our university and surrounding neighborhoods.

Writers Room is a diverse, intergenerational collective of students/alumni, faculty/staff, and neighborhood residents whose work demonstrates a desire for collaborative opportunities in our joint communities. A New Kind of House is created in partnership between Writers Room, ArtistYear and The Drexel Collection. Supported in part by Canon CSA and the Philadelphia Cultural Fund. The exhibition is curated by Lori Waselchuk.
